Active Site: In chymotrypsinogen, the substrate-binding pocket is inaccessible. Upon conversion to chymotrypsin, this pocket becomes exposed.
Serine: Serine 195 in chymotrypsinogen is non-acidic. Following conversion to chymotrypsin, Serine 195 becomes acidic.
Charge Relay: The charge relay system is non-functional in chymotrypsinogen but becomes active in chymotrypsin.
